چکیده

• Breast adenocarcinoma- brain endothelial de-adhesion dynamics is presented. Adenocarcinoma-derived exosomes alter pattern of endothelium. Intercellular mechanics might play important role in metastatic niche formation. Despite advances modern therapeutics, one the most feared complications cancer are metastases, which often cause life impairing profound neurological symptoms and premature death. adenocarcinoma among leading “sources” metastases. Since central nervous system lacks a classical lymphatic circulation, invading cells can reach parenchyma only through haematogenous routes must breach blood-brain barrier (BBB). The key step before transmigration highly regulated interface BBB establishment firm adhesion between tumor cell cerebral layer. Using atomic force microscopy, as high resolution spectrograph, direct measurements intercellular interactions was performed living confluent layer pre-treated with carcinoma cell-derived exosomes. By immobilization to an microscope’s cantilever, de-adhesions were directly measured by single spectroscopy (SCFS) at quasi-physiological conditions. De-adhesion strength characterized several different calculated parameters, involving aspects both membrane surface related factors. Our results indicate that lower case exosome compared non-treated controls. adenocarcinoma-derived have effect on

Thrombospondin modulates human breast adenocarcinoma cell adhesion to human vascular endothelial cells.

Thrombospondin (TSP), a M(r) 450,000 cytoadhesive glycoprotein, has been shown to potentiate tumor cell metastasis in mice by a mechanism that involves the hemostatic system of the host. In this study, the potential involvement of TSP in the interaction of human mammary adenocarcinoma MCF-7 cells with human umbilical vein endothelial cells (HUVECs) in culture was investigated. Using an ELISA, p...

Rat prostate adenocarcinoma cells disseminate to bone and adhere preferentially to bone marrow-derived endothelial cells.

Approximately 70% of patients with prostatic cancer develop bone metastases. Metastatic prostate adenocarcinomas are associated with high mortality rates and represent a leading cause of cancer-related deaths among males. To study the host-tumor interactions underlying the predilection of prostate cancer cells for skeletal bone, an experimental model was developed using rat Dunning carcinoma Ma...

Manipulation of hyaluronan synthase expression in prostate adenocarcinoma cells alters pericellular matrix retention and adhesion to bone marrow endothelial cells.

Prostate cancer metastasis to bone marrow involves initial adhesion of tumor cells to the bone marrow endothelium, followed by transmigration and proliferation within the marrow. Rapid, specific adhesion of highly metastatic prostate adenocarcinoma cells (PC3M-LN4) to bone marrow endothelial cell (BMEC) lines requires a pericellular hyaluronan (HA) matrix and correlates with dramatically up-reg...

Procyanidin B2 Cytotoxicity to MCF-7 Human Breast Adenocarcinoma Cells

Procyanidins have attracted some attention due to their demonstrated chemopreventive action, a relatively new and promising strategy to prevent cancer. Breast cancer is one of the leading causes of death in women worldwide and its treatment needs improvements. The aim of this work was to verify the procyanidin dimmer B2 cytotoxic effect to MCF-7 human breast cancer cells. MCF-7 cells were cultu...

Cardiomyocyte progenitor cell-derived exosomes stimulate migration of endothelial cells

Patients suffering from heart failure as a result of myocardial infarction are in need of heart transplantation. Unfortunately the number of donor hearts is very low and therefore new therapies are subject of investigation. Cell transplantation therapy upon myocardial infarction is a very promising strategy to replace the dead myocardium with viable cardiomyocytes, smooth muscle cells and endot...
